this is what i do when using 50mm,60mm or 105mm..

if your using it on M and your using full TTL mode on the flash u dont go by the metering system..the only time u go by that before u take a shot is if your using it in natural light and dont want to use a flash..then u would have to change various settings to get the histogram needle in the middle..

i use f/8-f/11 99% of the time
shutter speed 200 when using a flash
iso 100- iso200 when using a flash

when im not using a flash depending on the light i would knock up iso..400 max..

in natural light on a sunny day my typical settings with out using a flash are
iso400 f/11 shutter speed 125

that normally gets the light spot on..it will all depend on the light levels..lots of practice is the key,,

fill flash is when i have full ttl mode on the flash with the camera set on M.i would then change the settings until im 1 hole stop under exposed..your only using the flash for a tiny bit of light then..to fill areas that have shadows

if u want to shoot in natural light use tv mode and change iso to 400 your then need to change your f number..go with f/11...with tv mode the camara sets the shutter speed for u this way shooting in tv mode in natural light your always have the metering spot on..using the settings i said above will make sure the camara sets the shutter speed around 125-200 depending on light levels..this way u dont need a tripod then.
